From stem cells to Schwann cells

Abstract

Geïnduceerde pluripotente stamcellen, oftewel iPS-cellen, zijn stamcellen met soortgelijke eigenschappen als embryonale stamcellen. iPS-cellen kunnen gegenereerd worden uit somatische cellen door deze met specifieke pluripotentiefactoren te reprogrammeren tot een basale stamceltoestand. Onze resultaten laten zien dat neurale crest stamcellen, welke onder andere in de huid en de darm voorkomen, gereprogrammeerd kunnen worden tot iPS-cellen. Neurale crest stamcellen beschikken reeds over enkele van de specifieke pluripotentiefactoren; hun reprogrammering tot iPS cellen verloopt echter opmerkelijk genoeg niet efficiënter. Wij laten daarnaast zien dat iPS-cellen (gecreëerd uit muizenfibroblasten) efficiënt gedifferentieerd kunnen worden tot Schwanncellen, de ondersteunende gliacellen van het perifeer zenuwstelsel. Schwanncellen kunnen in combinatie met zenuwgeleiders een rol spelen in de behandeling van perifere zenuwletsels. Wij toonden aan dat Schwanncellen in een onderhuids geplaatste zenuwgeleider vervolgdkunnen worden met behulp van in vivo bioluminescent imaging. Tezamen kunnen deze bevindingen bijdragen aan de ontwikkeling van behandelmethodes voor behandeling van perifeer zenuwletsel met behulp van autologe (patiënt-eigen) Schwanncellen. Daarnaast beschrijven wij de aanwezigheid, verspreiding en mogelijke functie van een nog niet gekarakteriseerd eiwit, genaamd UGS148, in de hersenen. Onze resultaten tonen aan dat UGS148 niet alleen een rol speelt in de proliferatie van neurale stamcellen, maar ook in grote hoeveelheid aanwezig is in de hypothalamus, in het bijzonder in tanycyten. Tanycyten zijn stamcellen die zich kunnen ontwikkelen tot specifieke zenuwcellen die een rol spelen in onder andere voedselinname. Of UGS148 naast zijn rol in zenuwstamcellen daarmee ook een rol speelt in metabolismefuncties, blijft een interessante vraag voor nader onderzoek.; iPS cells are pluripotent stem cells that share important characteristics with embryonic stem cells. They can be created by the reprogramming of somatic cells, using specific pluripotency factors. Our results show that neural crest stem cells, which can be harvested from the skin and the gut, can be reprogrammed to become iPS cells. Although neural crest stem cells endogenously express some critical pluripotency factors, we show that this, surprisingly, does not facilitate their reprogramming towards iPS cells. Furthermore, our results show that mouse fibroblast-derived iPS cells can be efficiently differentiated towards Schwann cells, the glia cells of the peripheral nervous system. Schwann cells in combination with nerve guides can facilitate peripheral nerve repair. We have shown that Schwann cells in nerve guides can be visualized in vivo by means of bioluminescent imaging after subcutaneous implantation. Altogether, these results may contribute to the development of new modalities using autologous (patient-derived) Schwann cells for the treatment of peripheral nerve injuries.
